Description

This repository contains raw data that were used to draw conclusions and generate figures for the paper "Exploring the Aggregation Propensity of PHF6 Peptide Segments of the Tau Protein using Ion Mobility Mass Spectrometry Techniques". It includes readme files, raw data, and processed data. In this paper, we investigated the early steps of aggregation of the Ac-PHF6-NH2 peptide from tau protein using two types of ion mobility mass spectrometry techniques (trapped ion mobility - TIMS and traveling wave - TWIMS). We also compared the CCS values of the Ac-PHF6-NH2 peptide obtained by both methods. We also investigated the aggregation propensity of four different capped versions of this peptide, namely the Ac-PHF6-NH2, Ac-PHF6, PHF6-NH2 and uncapped PHF6 peptide segments. The aggregation propensity of the four PHF6 segments is confirmed using thioflavin T fluorescence assays and transmission electron microscopy. Finally, we present a new approach to improve peak assignment in ion mobility spectra using quadrupole selection on the TIMS instrument.
